以大蒜根尖分生区细胞为实验对象,试验了低温对有丝分裂细胞同步化的影响。采用不同低温处理为实验组,室温(19~21℃)处理为对照组。根尖细胞经培养后,光镜下观察、计数,并计算出各期细胞的有丝分裂指数,然后经过方差分析和q检验。结果表明:对照组与实验组有丝分裂指数差异有高度显著性或有显著性(P<0.01或P<0.05)。提示:低温处理能有效地积累分裂期细胞。
In order to test the effect of hypothermia on the synchronization of mitotic cells, the apical meristematic cells of garlic were used as experimental subjects. Different low-temperature treatment was used as experimental group, and the control group was treated at room temperature (19-21 ℃). After culturing the root tip cells, the cells were observed and counted under light microscope, and the mitotic index of each stage of cells was calculated, and then subjected to analysis of variance and q test. The results showed that there was a significant or significant difference in the mitotic index between the control group and the experimental group (P <0.01 or P <0.05). Tip: Cryogenic treatment can effectively divide cells.
