目的研究C_2mRNA及其蛋白在原发性肝细胞肝癌(HCC)中的表达及意义。方法用原位杂交检测C_2mRNA住21例HCC及其癌旁组织中的表达,ABC免疫组化法检测C_2蛋白在60例HCC及其42例癌旁组织中的表达,Western blot法检测C_2蛋白在HCC及其癌旁组织中的表达。结果 21例HCC及其癌旁组织中,C_2mRNA阳性率分别占23.8%(5/21)和85.7%(18/21),60例HCC及42例癌旁组织中,C_2蛋白阳性分别占27.3%(17/60)和83.3%(35/42),27例肝硬变中,C_2蛋白阳性率为77.8%(21/27)。χ~2检验:C_2mRNA及其蛋白在HCC癌旁组织中表达明显高于癌组织(P<0.001);C_2蛋白在肝硬变中的表达明显高于HCC癌组织(P<0.01);C_2的表达与患者年龄、HBsAg及AFP有明显关系(P<0.05);而与癌组织的分化程度、肿瘤大小、淋巴转移无关;Western blot与免疫组化结果一致。结论 C_2基因表达下调可能与HCC的发生、发展及早期诊断有关。
Objective To investigate the expression and significance of C 2 mRNA and its protein in primary hepatocellular carcinoma (HCC). Methods In situ hybridization was used to detect the expression of C 2 mRNA in 21 cases of HCC and its adjacent tissues. ABC immunohistochemistry was used to detect the expression of C 2 protein in 60 cases of HCC and its adjacent tissues. The C 2 protein was detected by Western blot. Expression in HCC and its adjacent tissues. Results The positive rate of C 2 mRNA was 23.8% (5/21) and 85.7% (18/21) in the 21 cases of HCC and paracancerous tissues respectively. In the 60 cases of HCC and 42 cases of paracancerous tissues, the positive rate of C 2 protein was 27.3% respectively. Among 17 cases (17/60) and 83.3% (35/42), in 27 cases of liver cirrhosis, the positive rate of C 2 protein was 77.8% (21/27). χ~2 test: The expression of C2 mRNA and its protein in HCC adjacent tissues was significantly higher than that in cancer tissues (P<0.001). The expression of C2 protein in liver cirrhosis was significantly higher than that in HCC tissues (P<0.01); C2 There was a significant relationship between the expression and the age of patients, HBsAg and AFP (P<0.05), but not with the degree of differentiation of cancer tissue, tumor size, lymph node metastasis; Western blot was consistent with the results of immunohistochemistry. Conclusion Down-regulation of C 2 gene expression may be related to the occurrence, development and early diagnosis of HCC.
