目的:观察研究加味四逆散对大鼠免疫性肝纤维化的防治效果,初步探索该方的防治机理。方法:采用人血清白蛋白(HSA)复制免疫性肝纤维化大鼠模型;设西药秋水仙碱组为对照;预防组在造模的第48天开始用加味四逆散给大鼠灌胃用药,共42天;治疗组在模型形成后(造模的第90天)开始用加味四逆散灌胃给药,共30天;用自动分析法检测大鼠血清ALT、AST、TP、ALB、GP含量;用光镜对大鼠肝脏进行病理组织学检查。结果:预防组和治疗组大鼠在应用加味四逆散后,血清ALT、AST、TBil明显降低;ALB升高,A/G值改善;肝脏组织病理学损害明显减轻,胶原沉积减少,肝细胞坏死得以修复,肝脏微循环有明显改善。与病理模型组比较有显务性差异(P<0.05)。与秋水仙碱组比较也有显著性差异(P>0.01)。结论:加味四逆散能有效的保护肝细胞,恢复肝功能,抑制星状细胞活性,减少肝内胶原蛋白的合成与沉积,促进胶原降解;具有肯定良好的预防和治疗肝纤维化作用,其预防效果优于治疗效果,并且明显优于秋水仙碱。
OBJECTIVE: To observe and study the preventive and therapeutic effects of Jiawei Sini San on immune hepatic fibrosis in rats, and to explore the prevention and treatment mechanism of this prescription. METHODS: Human serum albumin (HSA) was used to replicate the rat model of immune hepatic fibrosis; the colchicine group was used as the control; the prevention group was given intragastric administration to the rats on the 48th day after modeling. , A total of 42 days; treatment group after the formation of the model (the first 90 days of model) began to use gavage Sinisan intragastric administration for a total of 30 days; automatic analysis of rat serum ALT, AST, TP, ALB, The content of GP was detected by light microscopy. RESULTS: Serum ALT, AST, and TBil levels were significantly decreased in the prophylactic and treatment groups after administration of Modified Sini Powder. ALB increased, A/G value improved, histopathological damage was significantly reduced, collagen deposition was decreased, and hepatocytes were reduced. Necrosis can be repaired and the liver microcirculation is significantly improved. Compared with the pathological model group, there was a significant difference (P<0.05). There was also a significant difference from the colchicine group (P>0.01). Conclusion: Modified Sini Powder can effectively protect liver cells, restore liver function, inhibit the activity of stellate cells, reduce the synthesis and deposition of collagen in the liver, promote collagen degradation, and has a positive effect on the prevention and treatment of liver fibrosis. The preventive effect is better than the therapeutic effect and is significantly better than colchicine.
