目的探讨环境因素(如饮食)与遗传基因的相互作用对个体血脂性状的影响。方法以高脂乳剂喂养120只小鼠4w后,采用PCR-SSCP(聚合酶链反应单链构象多态性)方法检测小鼠脂蛋白脂酶(LPL)基因型对其血脂水平和部分器官性状的影响。结果高脂乳剂喂养4w后,120只小鼠中98只患有高脂血症,22只未患。在对所有小鼠基因组DNA进行PCR扩增和单链构象多态分析,发现在所扩增的220bp目的片段的14355处(GenBank accession No.NT039462)存在一个G/A的突变。相关性分析表明,BB基因型(A/A)小鼠的TC、TG、LDL、心/体重(H/W)、肝/体重(L/W)明显高于AA基因型(G/G)小鼠,差异显著(P<0.05);而AA基因型小鼠的HDL高于AB和BB基因型(P<0.05)。结论不同LPL基因型小鼠对高脂乳剂的敏感性存在一定的差异。
Objective To investigate the effects of environmental factors (such as diet) and genetic interactions on the lipid profile of individuals. Methods One hundred and twenty mice were fed with high-fat emulsion for 4 weeks. PCR-SSCP (single strand conformation polymorphism) method was used to detect the lipoprotein lipoprotein (LPL) Impact. Results After 4 weeks of high-fat emulsion feeding, 98 out of 120 mice had hyperlipidemia and 22 did not. After PCR amplification and single-stranded conformational polymorphism analysis of all mouse genomic DNA, a G / A mutation was found at 14355 of the amplified 220 bp fragment (GenBank accession No. NT039462). Correlation analysis showed that TC, TG, LDL, H / W and L / W in BB genotype mice were significantly higher than those in AA genotypes (G / G) (P <0.05). However, HDL of AA genotype mice was higher than that of AB and BB genotypes (P <0.05). Conclusion Different LPL genotype mice have different sensitivity to high fat emulsion.
