目的研究人羊膜间充质干细胞(hAMSCs)的免疫调节作用。方法采用兔双向混合淋巴细胞反应(MLR)体系,根据不同比例加入丝裂霉素处理过的hAMSCs,3H掺入法测定淋巴细胞增殖率并用ELISA法测定混合培养上清液中白细胞介素2(IL-2)和γ-干扰素(IFN-γ)的含量。结果 hAMSCs可以抑制兔混合淋巴细胞反应体系中淋巴细胞的增殖,抑制率与加入的hAMSCs数量成正比;同时hAMSCs可以减少混合淋巴细胞反应中细胞因子IL-2、IFN-γ的分泌。结论 羊膜间充质干细胞可以抑制兔混合淋巴细胞反应体系中淋巴细胞增殖。
Objective To study the immunomodulatory effects of human amniotic membrane-derived mesenchymal stem cells (hAMSCs). Methods Rabbit two - way mixed lymphocyte reaction (MLR) system was used. Mitomycin - treated hAMSCs were added according to different ratios. The proliferation rate of lymphocytes was determined by 3H - incorporation method. The interleukin - 2 IL-2) and interferon-γ (IFN-γ) content. Results hAMSCs could inhibit the proliferation of lymphocytes in rabbit mixed lymphocyte reaction system. The inhibition rate was proportional to the number of hAMSCs added. Meanwhile hAMSCs could reduce the secretion of cytokines IL-2 and IFN-γ in mixed lymphocyte reaction. Conclusion Amniotic mesenchymal stem cells can inhibit lymphocyte proliferation in rabbit mixed lymphocyte reaction system.
