目的探讨胰腺应激蛋白胰石蛋白/再生蛋白(PSP/reg)对胰腺星状细胞(PSC)的增殖、迁移、凋亡、胞外基质(ECM)合成的影响。方法分离纯化慢性胰腺炎患者纤维化区的PSC,基因重组胰腺应激蛋白PSP/reg,分别以10、100 ng/ml的终浓度对其进行干预。BrdU标记法检测PSC增殖;创伤愈合法观察PSC的迁移;原位缺口末端标记(TUNEL)技术检测PSC凋亡。细胞免疫荧光法观察细胞外基质(ECM)如胶原蛋白I(Col I)、纤连蛋白(FN)的生成。结果胰腺应激蛋白PSP/reg呈浓度依赖性地抑制PSC的增殖和迁移(P<0.05),而不影响PSC凋亡。免疫荧光结果显示Col I、FN生成减少。结论 胰腺应激蛋白PSP/reg在急慢性胰腺炎时显著升高。其抑制纤维化主要效应细胞PSC的活性,可能减少胰腺纤维化发生,在胰腺炎后的再生修复过程中发挥重要作用。
Objective To investigate the effects of pancreatic stress protein PSP / reg on the proliferation, migration, apoptosis and extracellular matrix (ECM) synthesis of pancreatic stellate cells (PSCs). Methods PSC and PSP / PSP were isolated and purified from fibrosis area of chronic pancreatitis patients, and were respectively treated with 10,100ng / ml of final concentration. Proliferation of PSC was detected by BrdU labeling; migration of PSC was observed by wound healing assay; apoptosis of PSC was detected by TUNEL. Cell immunofluorescence was used to observe the formation of extracellular matrix (ECM), such as collagen I (Col I) and fibronectin (FN). Results Pancreatic stress protein PSP / reg inhibited PSC proliferation and migration in a concentration-dependent manner (P <0.05) without affecting PSC apoptosis. Immunofluorescence results showed reduced Col I and FN production. Conclusion Pancreatic stress protein PSP / reg is significantly increased in acute and chronic pancreatitis. It inhibits the activity of PSC, a major effector of fibrosis, may reduce the occurrence of pancreatic fibrosis and plays an important role in the regeneration and repair of pancreatitis.
