为鉴别纯种铁观音和发生表型性状变化的铁观音茶树,本研究应用筛选出的11条ISSR引物对13份茶树样本进行分子标记分析。结果显示,11条引物共产生91个扩增位点,其中45个为多态性位点,多态性比率为49.45%。13份供试样本的遗传相似性系数分布在0.687~0.986之间,平均值0.805,聚类分析显示发生性状变化的铁观音茶树基本聚为一类,区别于纯种铁观音类群。依据扩增位点的特异性从中筛选出5条核心引物,对其扩增谱带编码后进行连锁组合构建了安溪铁观音茶树分子指纹图谱,依据图谱差异可鉴定性状不同的铁观音茶树。
In order to identify pure Tieguanyin and phenotypic trait changes Tieguanyin tea, the application of screening 11 ISSR primer 13 samples of tea tree molecular marker analysis. The results showed that 11 primers generated a total of 91 amplification sites, of which 45 were polymorphic sites, the rate of polymorphism was 49.45%. The genetic similarity coefficients of 13 samples ranged from 0.687 to 0.986, with an average of 0.805. Cluster analysis showed that Tieguanyin tea plants with the trait changes were basically clustered into one group, which was different from pure Guanyin group. According to the specificity of the amplification site, five core primers were screened out, and the amplified bands were encoded and linked to form the molecular fingerprint of tea tree of Anxi Tieguanyin. According to the differences of the spectra, the Tieguanyin tea tree with different traits could be identified.
