目的 探讨6 0 Coγ射线对不同p5 3基因功能状态的人淋巴性恶性肿瘤细胞株 82 2 6和Raji凋亡相关基因Bcl xL、Bax的作用及其分子机理。方法 分别运用流式细胞仪技术 (FACS)、DNA片段释放法、RT PCR和MTT法 ,分析了 82 2 6、Raji中Bcl xL、Bax转录相对表达与辐照诱导的细胞凋亡率及抑制细胞增殖的关系。结果 82 2 6和Raji在照射后Bcl xL、Bax转录表达均迅速上升 ,于 4~8h达到高峰 ,而后开始下降 ,于 2 4~ 48h恢复至辐照前水平 ;82 2 6中Bcl xL转录较Raji持续时间为长 ;且Bcl xL Bax的比率影响靶细胞辐照后的细胞凋亡和抑制细胞增殖。结论 肿瘤细胞株p5 3基因状态与辐射敏感性相关 ,两个等位基因突变的细胞株 82 2 6比一个等位基因突变的细胞株Raji更具辐射抗性 ,这种辐射抗性的不同与肿瘤细胞Bcl xL、Bax的表达及二者比率有关。
Objective To investigate the role and molecular mechanism of 60 Co γ-rays in human lymphatic malignant cell lines 82 26 and Raji apoptosis-related genes Bcl xL and Bax with different p53 gene expressions. Methods Flow cytometry (FACS), DNA fragment release assays, RT PCR, and MTT assays were used to analyze the relative expression of Bcl xL and Bax in Raji and the radiation-induced apoptosis rate and cell inhibition in Raji. RESULTS: The expression of BclxL and Bax in Raji increased rapidly after irradiation with Raji, which reached its peak at 4-8 hours, then began to decline, and returned to the pre-irradiation level at 24 to 48 hours. The transcription of Bcl-xL was stronger in 822.6. The Raji duration is long; and the ratio of BclxL Bax affects the apoptosis of the target cells after irradiation and inhibits cell proliferation. Conclusion The status of the p53 gene in tumor cell lines is related to radiation sensitivity. The two allele-mutant cell lines 82 2 6 are more radiation resistant than the allele-mutant cell line Raji.
