目的 研究内皮素 1(ET 1)对牛脑血管平滑肌细胞(CSMC)内源性ClC 3表达的影响。方法 采用培养的CSMC ,用免疫印迹 (westernblot)方法分析CSMCClC 3通道表达。结果 ①牛脑基底动脉、大脑中动脉、微动脉均有ClC 3蛋白表达 ,分子量约 10 5ku ,以微动脉表达最丰富 ,基底动脉表达最少 ;②培养的CSMC有内源性ClC 3蛋白表达 ,分子量约 95ku ;③ET 1能增加CSMCClC 3蛋白表达 ;④nifedipine、SK&F96 36 5能减少ET 1作用 ,环匹阿尼酸(cyclopiazonicacid ,CPA)浓度依赖性促进ClC 3蛋白表达 ,并能增强ET 1作用。结论 脑血管平滑肌细胞存在内源性ClC 3,ET 1可增强ClC 3表达 ,减少或增加胞浆内Ca2 + 浓度均可影响ET 1的作用
Objective To investigate the effect of endothelin 1 (ET 1) on the expression of endogenous ClC 3 in bovine cerebrovascular smooth muscle cells (CSMC). Methods Cultured CSMCs were used to analyze the expression of CSMCC13 channel by western blotting. Results ① The expression of ClC 3 protein in bovine basilar artery, middle cerebral artery and arterioles was about 105 ku, the most abundant in arterioles and the lowest in basilar artery. ② The cultured CSMC had endogenous ClC 3 protein expression, The molecular weight of about 95ku; ③ ET 1 can increase the expression of CSMCClC 3 protein; ④ nifedipine, SK & F96 36 5 can reduce ET 1 role, cyclopiazonic acid (cyclopiazonicacid, CPA) in a concentration-dependent manner to promote ClC 3 protein expression, and enhance the role of ET 1. Conclusions The existence of endogenous ClC 3 and ET 1 enhances the expression of ClC 3 in cerebrovascular smooth muscle cells. The effect of ET 1 can be influenced by decreasing or increasing the intracellular Ca 2+ concentration
