目的观察携带T-bet基因的重组腺相关病毒(rAAV-T-bet)对卵清蛋白(OVA)诱发的哮喘模型小鼠Blys水平的影响,探讨T-bet基因传递对哮喘的免疫调节作用。方法将40只Balb/c小鼠随机分为模型对照组(A组)、T-bet干预组(B组)、eGFP对照组(C组)、正常对照组(D组)。用ELISA法检测小鼠血清、肺泡灌洗液(BALF)和脾细胞上清液中Blys的水平。结果 A、C组BALF中Blys水平比正常对照组显著下降(P<0.05)。rAAV-T-bet干预后,BALF中Blys水平明显增高,并具有统计学意义(P<0.05)。各组小鼠血清和脾细胞上清液中Blys的水平变化类似BALF。结论应用rAAV-T-bet的哮喘小鼠气道炎症有明显减轻,其机制可能与表达的T-bet上调Thl/Th2比值,进而调整机体的免疫平衡有关。
Objective To observe the effect of rAAV-T-bet carrying T-bet gene on the level of Blys in ovariectomized (OVA) -induced asthmatic mice and to explore the immunoregulatory effect of T-bet gene delivery on asthma. Methods Forty Balb / c mice were randomly divided into three groups: model control group (A group), T-bet intervention group (B group), eGFP control group (C group) and normal control group (D group). The levels of Blys in mouse serum, BALF and splenocyte supernatants were detected by ELISA. Results The level of Blys in BALF in group A and group C was significantly lower than that in control group (P <0.05). After the intervention of rAAV-T-bet, the level of Blys in BALF was significantly increased, with statistical significance (P <0.05). The levels of Blys in serum and spleen cell supernatant of each group changed like BALF. Conclusions Airway inflammation in asthmatic mice treated with rAAV-T-bet was significantly reduced. The mechanism may be related to the up-regulation of Thl / Th2 ratio and the regulation of immune balance by T-bet expression.
