目的 :采用贝美格腹腔注射制作大鼠急性癫持续状态 ( SE)模型 ,观察海马神经元中 bcl- 2、bax、p5 3的表达。 方法 :通过将 SD大鼠脑海马切片行 bcl- 2、bax、p5 3的免疫组化染色 ,观察肿瘤相关基因在大鼠 SE模型海马中的作用。 结果 :bcl- 2在正常大鼠海马中不表达 ,发作高峰期在 CA 3区有弱表达 ;bax在正常大鼠海马中弱表达 ,在发作高峰期 CA3区强表达 ;p5 3(突变型 )在正常大鼠海马及 SE模型中均不表达。 结论 :急性癫持续状态可造成海马神经元的凋亡 ,其部位主要位于 CA 3、CA1区 ,而且肿瘤相关基因 bcl- 2、bax在 SE过程中发挥作用 ,而p5 3(突变型 )不发挥作用
OBJECTIVE: To establish a rat model of acute epileptic seizure (SE) by intraperitoneal injection of Beomes and observe the expression of bcl-2, bax and p5 3 in hippocampal neurons. Methods: The expression of bcl-2, bax and p5 3 in hippocampus of SD rats was detected by immunohistochemistry to observe the role of tumor-associated genes in hippocampus of rat SE model. Results: bcl-2 was not expressed in the hippocampus of normal rats. The expression of bcl-2 was weakly expressed in the CA 3 area at the peak of the onset. The expression of bax was weakly expressed in the hippocampus of normal rats, Not expressed in normal rat hippocampus and SE model. CONCLUSIONS: The acute status epilepticus can induce the apoptosis of hippocampal neurons. The sites are located in the CA 3 and CA 1 regions, and the tumor-associated genes bcl-2 and bax play a role in the SE process. However, p5 3 (mutant) Play a role
