目的观察中药制剂养精种子汤对缺氧雄性大鼠生精细胞凋亡的影响。方法36只雄性SD大鼠分为正常组、缺氧组、养精种子汤低剂量(10 g.kg-1.d-1)组和养精种子汤高剂量(35 g.kg-1.d-1)组(n=9)。用模拟海拔5 000 m的低压舱,造成大鼠生殖系统损伤模型,应用光镜、电镜、原位末端标记法(TUNEL)观察服用养精种子汤前后大鼠生精细胞凋亡情况。结果光镜下缺氧组示生精小管内生殖细胞排列紊乱,生精细胞数减少,生精上皮变薄,管腔内精子数明显减少,而养精种子汤高剂量组可见生精小管各级生精细胞发育正常,排列整齐,管腔内有大量成熟精子。电镜下缺氧组出现明显细胞凋亡情况,养精种子汤高剂量组明显改善。TUNEL法检测表明缺氧组与正常组、养精种子汤低剂量组和养精种子汤高剂量组相比生精细胞凋亡率明显增高(P<0.01)。结论养精种子汤能明显减少缺氧雄性大鼠的生精细胞凋亡。
Objective To observe the effect of Yangjing Seed Decoction, a traditional Chinese medicine preparation, on the apoptosis of spermatogenic cells in hypoxia male rats. Methods Thirty-six male Sprague-Dawley rats were divided into normal group, hypoxia group, high-seedling-seed soup low-dose group (10 g.kg-1.d-1) and high-dose Yangse seed soup (35 g.kg-1. D-1) groups (n=9). Using a simulated hypobaric chamber at an altitude of 5 000 m, a rat model of genital injury was created. Light microscopy, electron microscopy and in situ end labeling (TUNEL) were used to observe the apoptosis of spermatogenic cells in rats after administration of Yang Jing Decoction. Results Under light microscope, the hypoxia group showed that the arrangement of germ cells in the seminiferous tubules was disordered, the number of spermatogenic cells was reduced, the thickness of the seminiferous epithelium became thinner, and the number of spermatozoa in the lumen decreased significantly. In the high dose group of Yangjing seed soup, the spermatogenic tubules were observed in each group. Grades of spermatogenic cells are normal, well-arranged, and there are a large number of mature spermatozoa in the lumen. Under electron microscope, there was obvious cell apoptosis in the hypoxia group, and the high-dose group of Yangjing seed soup significantly improved. TUNEL assay showed that the apoptotic rate of spermatogenic cells was significantly higher in the hypoxia group than in the normal group, the low-dose Yangye seed decoction group and the high-dose Yangye seed decoction group (P<0.01). Conclusion Yangjing seed soup can significantly reduce the apoptosis of spermatogenic cells in hypoxia male rats.
