目的:研究延龄草根及根茎中的化学成分。方法:采用硅胶、聚酰胺、反相硅胶C18、Sephadex LH-20柱色谱和制备高效液相色谱等对其85%甲醇洗脱部位进行分离纯化。用MS、1D-NMR及2D-NMR等波谱学方法鉴定化合物的结构。结果:从延龄草的85%甲醇洗脱部位中分离鉴定了10个甾体皂苷类化合物,分别鉴定为:薯蓣皂苷元-3-O-α-L-吡喃鼠李糖基(1→4)-α-L-吡喃鼠李糖基(1→4)-[α-L-吡喃鼠李糖基(1→2)]-β-D-吡喃葡萄糖苷(1)、trikamsteroside E(2)、deoxytrikamsteroside E(3)、(23S,24S,25S)-螺甾-5-烯-1β,3β,21,23,24-五羟基-1β-O-α-L-吡喃鼠李糖基-(1→2)-[O-β-D-吡喃木糖基(1→3)]-O-α-L-吡喃阿拉伯糖苷(4)、trikamsteroside B(5)、27-hydroxytrikamsteroside B(6)、3-乙酰基-偏诺皂苷元(7)、dioscoreanoside I(8)、sileneoside G(9)、intergristerone B(10)。结论:其中,化合物8~10为首次从该属植物中分离得到,化合物1、3、4、6、7为首次从该植物中分离得到。
Objective: To study the chemical constituents of Trilliums roots and rhizomes. Methods: The 85% methanol elution was separated and purified by silica gel, polyamide, reversed-phase silica gel C18, Sephadex LH-20 column chromatography and preparative high performance liquid chromatography. The structures of the compounds were identified by spectroscopic methods such as MS, 1D-NMR and 2D-NMR. Results: Ten steroidal saponins were isolated from 85% methanol eluting fractions of Trillium chinense and identified as: diosgenin -3-O-α-L-rhamnopyranosyl (1 → 4) -alpha-L-rhamnopyranosyl (1 → 4) - [α-L-rhamnopyranosyl (1 → 2)] - β-D-glucopyranoside (1), trikamsteroside E (2), deoxytrikamsteroside E (3), (23S, 24S, 25S) -spirost-5-en-1β, 3β, 21,23,24-pentahydroxy-1β-O-α-L- (1 → 2) - [O-β-D-xylopyranosyl (1 → 3)] - O-α-L-arabinopyranoside (4), trikamsteroside B -hydroxytrikamsteroside B (6), 3-acetyl-sennoside (7), dioscoreanoside I (8), sileneoside G (9), intergristerone B (10). Conclusion: Compounds 8 to 10 were isolated from this genus for the first time. Compounds 1, 3, 4, 6 and 7 were isolated from this plant for the first time.
