目的:研究狼紫草水提物对小鼠皮肤溃疡组织环氧酶-2(cyclooxygenase-2,COX-2)表达的影响。方法:煎煮法制备狼紫草水提物,醋酸化学刺激法制备小鼠体表溃疡模型。分别采用Western blotting和RT-PCR法测定局部皮肤溃疡组织中环氧酶-2蛋白及其mRNA表达情况。结果:正常小鼠皮肤组织中几乎无COX-2表达,模型组小鼠体表溃疡组织中COX-2蛋白及其mRNA的表达均显著增加,应用高剂量(40g.kg-1)和中剂量(20g.kg-1)狼紫草水提物治疗后,二者表达水平明显降低。结论:狼紫草水提物对COX-2有明显的干预作用,可预见其抗炎镇痛作用有较好应用前景。
OBJECTIVE: To study the effect of water extract of Wolfowder on the expression of cyclooxygenase-2 (COX-2) in skin ulcer of mice. Methods: The decoction method was used to prepare the water extract of Zanthoxylum, and the acetic acid chemical stimulation method was used to prepare the mouse surface ulcer model. Western blotting and RT-PCR were used to determine the expression of cyclooxygenase-2 protein and its mRNA in local skin ulcers. Results: There was almost no COX-2 expression in the skin of normal mice. The expression of COX-2 protein and mRNA in the ulcer tissue of the model group was significantly increased. The high dose (40g.kg-1) (20g.kg-1) wolfberry water extract treatment, both significantly reduced the expression level. Conclusion: The water extract of Wolfberry grass has a significant intervention on COX-2, and its anti-inflammatory and analgesic effects can be forecasted to have a good application prospect.
