目的 研究u PA及其抑制物PAI 1在大鼠thy 1肾炎中的表达及意义 ,并探讨肝素治疗对其表达的影响。方法 应用ABC免疫酶标法及图像分析技术 ,观察对照组、肾炎组和肝素组动物模型肾小球u PA和PAI 1的表达情况 ,并与光镜下肾小球细胞计数作相关性分析。结果 肝素组动物在其治疗后 7、14、2 1d时 ,肾小球细胞数明显减少 ,与肾炎组比较P值分别小于 0 .0 5或 0 .0 1:肾炎组、肝素组肾小球u PA和PAI 1的表达均高于对照组 ;于实验 3、7、14和 2 1d时 ,肾炎组动物的肾小球细胞数增多与u PA和PAI 1的表达呈正相关 (P <0 .0 5或 0 .0 1) ;肝素组于实验 3、7d时 ,肾小球细胞数的减少与PAI 1表达降低有关 (P <0 .0 5 )。结论 大鼠thy 1肾炎的肾小球细胞数增多与其u PA和PAI 1表达程度呈正相关 ;肝素治疗可减轻大鼠thy 1肾炎肾小球细胞增生 ,其作用机制可能与病程早期对肾小球PAI 1表达的抑制有关
Objective To study the expression and significance of u PA and its inhibitor PAI 1 in rat thy nephritis and to investigate the effect of heparin treatment on its expression. Methods The expressions of u PA and PAI 1 in the glomerulus of the control group, nephritis group and heparin group were observed by ABC immunohistochemistry and image analysis, and the correlation was analyzed with the count of glomerular cells under light microscope. Results Heparin group at 7,14,2 1d after treatment, the number of glomerular cells was significantly reduced, compared with the nephritis group P values were less than 0.05 or 0. 01: nephritis group, heparin group glomerular u PA and PAI 1 were higher than that of the control group. At 3, 7, 14 and 21 days, the number of glomerular cells in nephritis group was positively correlated with the expression of u PA and PAI 1 (P <0. 0 5 or 0 .0 1). In the heparin group, the decrease of glomerular cell number was associated with the decrease of PAI 1 expression at 3 and 7 days (P <0.05). Conclusions The increase of glomerular cells in thy1 nephritis rats is positively correlated with the expression of u PA and PAI1. Heparin treatment can reduce the glomerular hyperplasia in rats with thy1 nephritis. The mechanism may be related to the earlier stage of glomeruli Inhibition of PAI 1 expression
