目的:研究1-甲基-3-异丁基黄嘌呤(MIBX)对去除生长因子(aFGF和血清)诱导的血管内皮细胞凋亡的影响.方法:通过细胞存活率的分析,荧光显微技术和DNA凝胶电泳等方法,检测MIBX对细胞凋亡的影响.结果:用25-200μmol/L的MIBX处理培养在无aFGF和血清的培养液中的血管内皮细胞,50-200μmol/L的MIBX在处理6h明显抑制了凋亡小体的形成和DNA的片断化.但是同样浓度的MIBX处理细胞12h以后,处理组和对照组之间无明显差别.结论:MIBX延迟去除aFGF和血清诱导的血管内皮细胞凋亡.
Objective: To investigate the effect of 1-methyl-3-isobutylxanthine (MIBX) on the apoptosis of vascular endothelial cells induced by the removal of growth factors (aFGF and serum) .Methods: The cell viability was analyzed by fluorescence microscopy And DNA gel electrophoresis were used to detect the effect of MIBX on apoptosis.Results: The vascular endothelial cells cultured in culture medium without aFGF and serum were treated with 25-200 μmol / L MIBX, and 50-200 μmol / L MIBX 6 h treatment significantly inhibited the formation of apoptotic bodies and DNA fragmentation.However, after treated with the same concentration of MIBX for 12 h, there was no significant difference between the treatment group and the control group.Conclusion: MIBX delayed the removal of aFGF and serum-induced blood vessels Endothelial cell apoptosis.
