目的:探讨体外循环心脏缺血一再灌注对人心房肌一氧化氮合成酶的影响。方法:用免疫组织化学结合计算机图像分析技术,检测在心脏缺血一再灌注前后11例病人心房肌肉一氧化氮合成酶的三种同功异构酶一神经性一氧化氮合成酶、诱导性一氧化氮合成酶和内应性一氧化氮合成酶含量的变化。结果:心脏缺血一再灌注后神经性一氧化氮合成酶反应产物的平均灰度值明显减少,内皮性一氧化氮合成酶者明显增加,而诱导性一氧化氮合成酶者无明显改变。结论:心脏缺血一再灌注引起人心房肌神经性一氧化氮合成酶表达增加、内皮性一氧化氮合成酶表达减少,而对诱导性一氧化氮合成酶无明显影响。心脏缺血一再灌注后心脏功能失调可能与心房肌以上变化有关。
Objective: To investigate the effect of cardiopulmonary bypass with reperfusion on the activity of nitric oxide synthase in human atrial myocardium. Methods: Immunohistochemistry and computer image analysis were used to detect the expression of three isoforms of nitric oxide synthase, atrial nitric oxide synthase, in 11 patients with myocardial ischemia and reperfusion Changes of nitric oxide synthase and intrinsic nitric oxide synthase. Results: After reperfusion of ischemia, the average gray value of NOS product was significantly decreased, while that of endothelial nitric oxide synthase was significantly increased, while that of inducible nitric oxide synthase was unchanged. CONCLUSION: The reperfusion of cardiac ischemia causes the expression of neuronal nitric oxide synthase in human atrium to increase, the expression of endothelial nitric oxide synthase decreases, but has no effect on inducible nitric oxide synthase. Cardiac ischemia reperfusion after cardiac dysfunction may be related to changes in atrial muscle above.
