目的观察大承气汤对内毒素血症大鼠血清内毒素和结肠cAMP、cGMP的影响。方法 SD大鼠随机分为正常组,模型组,大承气汤高剂量组(5 g/kg·q12 h)、中剂量组(2.5 g/kg·q12h)、低剂量组(1.25 g/kg·q12h)。正常组行假手术,其余各组均盲肠结扎穿孔术致内毒素血症模型,给药5次后,取样检测血清内毒素和结肠cAMP、cGMP水平。结果大承气汤高、中剂量组血清内毒素、cAMP、cAMP/cGMP均明显降低,与模型组比较,有显著性差异(P<0.01);高剂量组cGMP明显升高,与模型组比较,有显著性差异(P<0.01);高剂量组在降低血清内毒素、cAMP、cAMP/cGMP比值,升高cGMP水平方面优于低剂量组(P<0.05)。结论大承气汤具有急下存阴功效,其机理可能与降低血清内毒素、cAMP及cAMP/cGMP比值有关。
Objective To observe the effects of Dachengqi Decoction on serum endotoxin and cAMP and cGMP in rats with endotoxemia. Methods SD rats were randomly divided into high dose group (5 g / kg · q12 h), middle dose group (2.5 g / kg · q12h), low dose group (1.25 g / kg · Q12h). The rats in the normal group were sham-operated, and the other groups were induced by cecal ligation and perforation. After five times of administration, samples were taken to detect serum endotoxin and cAMP and cGMP levels in the colon. Results Serum endotoxin, cAMP and cAMP / cGMP in Dachengqi Decoction group were significantly lower than those in model group (P <0.01), while cGMP in high dose group was significantly higher than that in model group (P <0.01). The high dose group was superior to the low dose group (P <0.05) in reducing the serum endotoxin, cAMP, cAMP / cGMP ratio and increasing cGMP level. Conclusions Dachengqi Decoction has the effect of urgency to keep the yin, and its mechanism may be related to the decrease of serum endotoxin, cAMP and cAMP / cGMP ratio.
