目的:观察三氧化二砷(As_2O_3)对人胆囊癌细胞体内外生长的抑制作用及对细胞周期的影响。方法:不同浓度As_2O_3作用于人胆囊癌GBC细胞,用MTT法检测细胞生长情况,流式细胞仪检测细胞周期分布;20只BALB/c裸鼠皮下接种GBC细胞,随机分为0.9%氯化钠(NS)组(n=10)和As_2O_3组(n=10)。腹腔注射0.9%氯化钠0.2ml·只~1(NS组)或As_2O_3 10 mg·kg~(-1)(As_2O_3组),每周2次,连续7周,测量肿瘤的体积。结果:As_2O_3能抑制体内、外GBC细胞生长.体外实验中As_2O_3作用呈时间、剂量效应关系;流式细胞仪检测发现As_2O_3可将增殖过程中的GBC细胞阻滞于G1期。结论:As_2O_3能明显抑制体内外人胆囊癌GBC细胞的生长及引起G1期阻滞。
Objective: To observe the inhibitory effect of arsenic trioxide (As_2O_3) on the growth of human gallbladder carcinoma cells in vitro and in vivo and its effect on cell cycle. Methods: The effect of different concentrations of As_2O_3 on human gallbladder carcinoma GBC cells was detected by MTT assay. The cell cycle distribution was detected by flow cytometry. Twenty BALB / c nude mice were inoculated subcutaneously with GBC cells and were randomly divided into 0.9% sodium chloride (NS) group (n = 10) and As 2 O 3 group (n = 10). The volume of tumor was measured by intraperitoneal injection of 0.9% sodium chloride (0.2ml · · min -1) or As 2 O 3 10 mg · kg -1 (As 2 O 3) twice a week for 7 weeks. RESULTS: As 2 O 3 inhibited the growth of GBC cells in vivo and in vitro, and the effect of As 2 O 3 in vitro was time and dose-dependent. As 2 O 3 could inhibit the proliferation of GBC cells in G1 phase by flow cytometry. Conclusion: As 2 O 3 can significantly inhibit the growth of GBC cells in vitro and in vivo and cause G1 arrest.
