目的探讨职业性苯所致慢性粒细胞白血病患者血小板线粒体三磷酸腺苷合成酶6亚基(ATPase-6)基因变化情况。方法采用方便抽样方法,选取5例职业性苯所致慢性粒细胞白血病患者作为病例组,以21名无职业性苯接触史的健康人员为对照组,抽取2组人员肘静脉血,检测血小板计数,采用荧光定量聚合酶链式反应法检测血小板线粒体DNA中ATPase-6基因水平。结果病例组人员血小板计数低于对照组[(135.2±32.6)×10~9个/L vs(239.7±77.5)×10~9个/L,P<0.05],ATPase-6基因/血小板比值高于对照组[中位数:3.43×10~(-4)copies/个vs1.87×10~(-4)copies/个,P<0.01]。病例组人员血小板线粒体ATPase-6基因水平与对照组比较,差异无统计学意义[中位数:4.83×10~7copies/L vs 3.24×10~7copies/L,P>0.05]。结论苯所致慢性粒细胞白血病患者ATPase-6基因/血小板比值较ATPase-6基因水平变化敏感。
Objective To investigate the changes of platelet mitochondrial ATPase-6 (ATPase-6) gene in patients with benzene-induced chronic myeloid leukemia. Methods Using a convenient sampling method, 5 cases of occupational benzene-induced chronic myelogenous leukemia were selected as the case group. 21 healthy workers without occupational benzene exposure were selected as the control group. The elbow venous blood was collected from 2 groups and the platelet count , The level of ATPase-6 gene in platelet mitochondrial DNA was detected by fluorescence quantitative polymerase chain reaction. Results The platelet counts in case group were lower than those in control group [(135.2 ± 32.6) × 10-9 / L vs (239.7 ± 77.5) × 10-9 / L, P <0.05) In the control group [median: 3.43 × 10 ~ (-4) copies / one vs1.87 × 10 ~ (-4) copies / one, P <0.01]. There was no significant difference in platelet mitochondrial ATPase-6 gene level between the two groups (median: 4.83 × 10 ~ 7copies / L vs 3.24 × 10 ~ 7copies / L, P> 0.05). Conclusion The ATPase-6 gene / platelet ratio in patients with benzene-induced chronic myeloid leukemia is more sensitive than that of ATPase-6 gene.
