探讨磷脂酶A2 (PLA2 )活性的变化对中性粒细胞凋亡的影响。方法 :采用光镜、电镜、DNA电泳和FACS等方法 ,分析了磷脂酶A2 内外源性激活剂和抑制剂对大鼠中性粒细胞凋亡的影响。结果 :脂多糖 (LPS1μg/ml)和钙离子载体 (A2 31871μmol/L)等PLA2 激动剂分别在共同培养 6h和 12h之内促进中性粒细胞凋亡 ,而PLA2 阻断剂 4 溴苯甲酰溴甲烷 (p BPB 0 .0 1~ 1mmol/L)可明显减弱上述激动剂的促凋亡作用。但另一种PLA2 非特异性抑制剂氯喹和血小板激活因子受体拮抗剂SR2 74 17A则无上述相同的作用。结论 :结果表明PLA2 激活可促进中性粒细胞凋亡过程 ,但该过程可能同PLA2 的代谢产物血小板激活因子的直接作用无关。
To investigate the effect of phospholipase A2 (PLA2) activity on neutrophil apoptosis. Methods: The effects of phospholipase A2 extrinsic activators and inhibitors on apoptosis of neutrophils in rats were analyzed by light microscope, electron microscope, DNA electrophoresis and FACS. RESULTS: PLA2 agonists such as lipopolysaccharide (LPS 1μg / ml) and calcium ionophore (A2 31871μmol / L) promoted the apoptosis of neutrophils within 6h and 12h, respectively. PLA2 antagonist 4-bromobenzoyl Methyl bromide (pBPB 0 .0 1 ~ 1mmol / L) can significantly reduce the above-mentioned agonist of apoptosis. However, another non-specific inhibitor of PLA2 chloroquine and platelet activating factor receptor antagonist SR2 74 17A do not have the same effect. CONCLUSION: The results show that PLA2 activation can promote neutrophil apoptosis, but this process may not be related to the direct effect of PLA2 metabolite platelet activating factor.
