鉴定人胃癌细胞系BGC-823中的SP(side population)细胞,并进一步研究SP细胞对化疗药物5-氟尿嘧啶的耐受性。利用Hoechst33342对细胞染色,通过流式细胞仪检测及分选Hoechst低染的SP细胞;同样的方法检测5-氟尿嘧啶处理后的细胞中SP细胞比例的变化;MTT法检测分选后的SP与非SP细胞的抗药性差异。结果表明,人胃癌细胞系BGC-823中存在4%左右的SP细胞;细胞经过5-氟尿嘧啶处理后,SP细胞的比例显著升高;体外正常培养条件下SP细胞并不具备增殖优势,但分选后的SP细胞较非SP细胞显示出了更强的耐药存活能力。
SP (side population) cells in human gastric cancer cell line BGC-823 were identified and the tolerance of SP cells to 5-fluorouracil chemotherapy was further investigated. The cells were stained with Hoechst 33342, and Hoechst stained SP cells were detected and sorted by flow cytometry. The same method was used to detect the proportion of SP cells in 5-fluorouracil-treated cells. MTT method was used to detect the SP and non- SP cell resistance differences. The results showed that about 4% of SP cells were present in human gastric cancer cell line BGC-823. The proportion of SP cells was significantly increased after treated with 5-fluorouracil. SP cells did not have the proliferation advantage under normal culture conditions, The selected SP cells showed stronger drug-resistant survival than non-SP cells.
