目的 :研究ets l反义寡核苷酸对胃癌细胞株SGC 790 1体外侵袭力的影响。方法 :应用ets l反义、正义寡核苷酸转染SGC 790 1细胞 ,并设PBS对照组。转染后应用逆转录聚合酶链式反应检测ets lmRNA的变化 ,应用克隆形成试验检测细胞增殖活性的变化 ,Transwell小室检测侵袭能力的变化。结果 :转染ets l反义寡核苷酸后 ,ets lmRNA表达降低 ,反义组形成克隆数目 (2 4 2± 4 8)较正义组、对照组减少 (47 6± 8 1,4 4 3± 7 6 ,P <0 0 1) ,侵过小室滤膜细胞数目减少 (97 1± 11 1,198 7± 18 3,2 0 5 8± 2 2 2 ,P <0 0 1)。结论 :ets l反义寡核苷酸能够抑制胃癌细胞株SGC 790 1的体外侵袭力。
Objective: To investigate the effect of ets l antisense oligonucleotide on the invasiveness of gastric cancer cell line SGC 790 1 in vitro. METHODS: SGC7901 cells were transfected with ets 1 antisense and sense oligonucleotide and PBS control group was established. The transcriptase polymerase chain reaction was used to detect the changes of ets lmRNA after transfection. The changes of cell proliferative activity were detected by clonogenic assay and the changes of invasion ability were detected by Transwell chamber. Results: The expression of ets lmRNA decreased after transfection with etsl antisense oligonucleotide. The number of antisense clones was (4242 ± 48) more than that of the sense group and 476 ± 8 1,443 ± 7 6, P <0 01). The number of cell invasion cells decreased (97 1 ± 11 1,198 7 ± 18 3,2 0 58 ± 2 2 2, P 0 01). Conclusion: ets l antisense oligonucleotide can inhibit gastric cancer cell line SGC 790 1 in vitro invasiveness.
