观察二烯丙基二硫(DADS)对人白血病 HL-60细胞增殖和钙网蛋白(CRT)表达的影响.DADS (1.25 mg/L)处理HL-60细胞24、48和72 h,siRNA抑制CRT的表达,采用MTT法检测细胞活力,实时定量PCR和Western blot检测CRT的表达.与对照组比较,DADS处理24、48和72 h组HL-6细胞的增殖水平均显著性降低,呈现时间依赖性(均P<0.05).与对照组比较,siRNA瞬时转染显著降低了CRT mRNA的表达(P<0.05),也显著降低了细胞的增殖水平(P<0.05),与DADS具有协同效应.DADS抑制了HL-60细胞增殖,其机制可能与下调钙网蛋白的表达有关.“,”To observe the effect of Diallyl disulfide (DADS) on the cell proliferation and expression of calreticulin (CRT) in human leukemia HL-60 cells.HL-60 cells were incubated with DADS (1.25mg/L) for 24,48 and 72 h.Expres-sion of CRT was inhibited by siRNA.Cell viability was tested by MTT assay.Expression of CRT was test by real-time quanti-tative PCR and Western bolt.Compared with the control group,the proliferation of HL-60 cells was significantly decreased in time dependent manner after treatment with DADS for 24,48 and 72 h.Compared with the control group,Expression of CRT was significantly decreased by transient transfection of siRNA of CRT.There was a synergistic effect between DADS and siR-NA of CRT on expression of CRT.Compared with the control group,the proliferation of HL-60 cells was significantly de-creased by siRNA of CRT.There was a synergistic effect between DADS and siRNA of CRT on the proliferation of HL-60 cells.DADS inhibits cell proliferation in human leukemia HL-60 cells,which mechanism may be related with down-regulation of CRT expression.
