目的:研究真姬菇多糖分子特性及生理活性,充分开发真姬菇资源。方法:沸水浸提真姬菇子实体多糖(HMP),用高效液相色谱(HPLC)及高效凝胶尺寸排阻色谱-十八角激光散色仪-示差检测仪联用(HPSEC-MALLS-RI)技术进行单糖含量和平均分子质量等分子特性的研究。选用WST-1法、Griess法和酶联免疫法(ELISA)研究HMP对人胃腺癌细胞(AGS)、结肠腺癌细胞(DLD-1)、子宫颈癌细胞(HeLa)的体外抑制作用及对小鼠巨噬细胞Raw264.7的免疫调节作用。结果:HMP由葡萄糖(89.2%)和半乳糖(10.8%)组成,平均分子质量分布于14770.0×104、1382.0×104、140.9×104、70.2×104u。HMP虽然对AGS、DLD-1、HeLa没有明显的体外抑制作用,但可以促进Raw264.7的增值和剂量依赖性地提高巨噬细胞中一氧化氮(NO)和前列腺素(PGE2)的生成量。结论:HMP具有免疫调节活性,可以作为免疫调节剂应用于保健品和医药行业。
Objective: To study the molecular characteristics and physiological activities of Pleurotus eryngii polysaccharides and to develop the resources of Pleurotus ostreatus. Methods: HMP was extracted by boiling water and characterized by high performance liquid chromatography (HPLC) and high performance gel exclusion chromatography (HPSEC-MALLS- RI) technology for monosaccharide content and average molecular weight and other molecular properties of the study. The inhibitory effects of HMP on human gastric adenocarcinoma cells (AGS), colon adenocarcinoma cells (DLD-1) and cervical cancer cells (HeLa) were studied by WST-1 assay, Griess assay and enzyme-linked immunosorbent assay (ELISA) Immune regulation of mouse macrophage Raw264.7. Results: HMP consisted of glucose (89.2%) and galactose (10.8%) with average molecular mass distribution at 14770.0 × 104,1382.0 × 104,140.9 × 104,70.2 × 104u. Although HMP had no obvious in vitro inhibition on AGS, DLD-1 and HeLa, HMP could promote the proliferation of Raw264.7 and increase the production of nitric oxide (NO) and prostaglandin (PGE2) in macrophages in a dose-dependent manner . Conclusion: HMP has immunomodulatory activity and can be used as an immunomodulator in health products and pharmaceutical industry.
