目的:探讨益骨口服液含药血清对成骨细胞增殖、分化及矿化功能的影响。方法:取SD大鼠40只,雌雄各半。将大鼠分为A组(高剂量组)、B组(中剂量组)、C组(低剂量组)、D组(空白组),E组(对照组)。按照不同的剂量灌胃。灌胃10d后,制备含药血清;将从乳鼠颅骨取材的成骨细胞培养于含药血清培养液中,然后应用MTT法、硝基苯磷酸盐法及茜素红染色方法观察体外培养成骨细胞、碱性磷酸酶及矿化结节的变化。结果:经过10d益骨口服液灌胃后采血制备的含药血清明显增加了成骨细胞数量(P<0.05或P<0.01)、碱性磷酸酶的表达水平及矿化结节的形成明显。结论:证实了益骨口服液含药血清可以促进成骨细胞增殖、分化及矿化。
Objective: To investigate the effects of Yi Gu Oral Liquid-containing serum on the proliferation, differentiation and mineralization of osteoblasts. Methods: 40 SD rats were chosen, male and female were divided into two groups. Rats were divided into group A (high dose group), group B (medium dose group), group C (low dose group), group D (blank group) and group E (control group). Gavage at different doses. Osteoblasts drawn from the suckling rat skull were cultured in the drug-containing serum culture medium after 10 days of intragastric administration, and then the serum containing medicine was prepared. Then MTT assay, nitrobenzene phosphate method and alizarin red staining were used to observe the in vitro culture Changes of osteocytes, alkaline phosphatase and mineralized nodules. Results: After 10 days of oral administration, the drug-containing serum prepared by gavage significantly increased the number of osteoblasts (P <0.05 or P <0.01). The expression of alkaline phosphatase and the formation of mineralized nodules were obvious. Conclusion: It is confirmed that the medicine serum of Yi Gu Oral Liquid can promote the proliferation, differentiation and mineralization of osteoblasts.
