观察巨噬细胞源趋化因子(macrophage-derived chemokine,MDC)联合不可分型流感嗜血杆菌重组P6蛋白免疫小鼠的免疫效果,探讨MDC的佐剂效应。将50只BALB/c小鼠随机分为5组:P6重组蛋白+MDC,P6重组蛋白,MDC组,P6重组蛋白+IL-2组及PBS对照组,分别于第0 d、14 d、28 d经腹腔免疫3次,末次免疫后14 d,取小鼠血、脾标本,ELISA法检测血清中抗体IgG水平,并分析其脾淋巴细胞IL-10和IFN-γ的产生水平。CCK-8法检测各组脾淋巴细胞增殖活性。结果是小鼠血清IgG水平,P6重组蛋白联合MDC组高于PBS对照组、MDC组、P6重组蛋白+IL-2组及P6重组蛋白组(P<0.05)。P6重组蛋白联合MDC组小鼠特异性脾淋巴细胞刺激指数及其所产生的IL-10、IFN-γ水平均高于PBS对照组、MDC组及P6重组蛋白组(P<0.05)。因此MDC联合P6重组蛋白诱导小鼠的免疫应答优于P6重组蛋白单独免疫,MDC可以作为不可分型流感嗜血杆菌P6重组蛋白的佐剂。
To observe the immune effect of macrophage-derived chemokine (MDC) combined with recombinant Haemophilus influenzae P6 protein immunized mice to explore the adjuvant effect of MDC. 50 BALB / c mice were randomly divided into 5 groups: P6 recombinant protein + MDC, P6 recombinant protein, MDC group, P6 recombinant protein + IL-2 group and PBS control group, d were immunized intraperitoneally for 3 times. At 14 days after the last immunization, blood and spleen samples from mice were collected. Serum antibody IgG levels were detected by ELISA. The levels of IL-10 and IFN-γ in spleen lymphocytes were analyzed. CCK-8 method was used to detect the proliferation activity of splenic lymphocytes in each group. The results were serum IgG levels of mice, P6 recombinant protein combined with MDC group was higher than PBS control group, MDC group, P6 recombinant protein + IL-2 group and P6 recombinant protein group (P <0.05). The specific spleen lymphocyte stimulating index and the level of IL-10 and IFN-γ produced by P6 recombinant protein combined with MDC group were higher than those of PBS control group, MDC group and P6 recombinant protein group (P <0.05). Therefore, MDC combined P6 recombinant protein induced immune response in mice is superior to P6 recombinant protein alone immunization, MDC can be used as an adjuvant of non-typeable Haemophilus influenzae P6 recombinant protein.
