目的探讨 IL-4受体基因 Ile50Val位点多态性和哮喘易感性及时血浆总 IgE、可溶性 IL-4受体水平的影响。方法用等位基因特异性 PCR 法分析位点多态性,ELISA 法测定血浆总 IgE 和可溶性 IL-4R 的浓度。结果对照组和哮喘组中,Ile/Val 基因型频率分布差异有显著性 x~2=18.94,P<0.01,两组中两等位基因的分布差异有显著性 x~2=20.88,P<0.01,变异等位基因携带者惠哮喘的风险高于未携带者,OR=2.64,P<0.01。在对照组和哮喘组中 Ile/Ile 基因型血浆总 IgE 和可溶性 IL-4R 的浓度明显高于 Val/Val 基因型,同基因型哮喘组血浆总 IgE 和可溶性 IL-4R 的浓度明显高于对照组。结论 IL-4受体基因 Ile50Val 位点多态性和哮喘相关,并影响血浆总 IgE、可溶性 IL-4受体水平。
Objective To investigate the influence of IL-4 receptor gene Ile50Val polymorphism and susceptibility to asthma on the levels of total serum IgE and soluble IL-4 receptor. Methods Allele-specific PCR was used to analyze the polymorphism of the loci. The concentrations of total IgE and soluble IL-4R in plasma were determined by ELISA. Results The frequency distribution of Ile / Val genotype in control group and asthma group was significantly different from that in control group (P <0.01). There was significant difference in the distribution of allele between two groups (P < 0.01, mutation allele carriers benefit asthma is higher than the risk of non-carriers, OR = 2.64, P <0.01. The concentrations of total IgE and soluble IL-4R in plasma of the Ile / Ile genotypes were significantly higher in the control group and the asthma group than those in the Val / Val genotype. The concentrations of total IgE and soluble IL-4R in the Ile / Ile genotype were significantly higher than those in the control group. Conclusion The IL-4 receptor gene Ile50Val polymorphism is associated with asthma and affects the levels of total IgE and soluble IL-4 receptor in plasma.
