采用流式细胞术对昆明小鼠接受75mGy和2GyX射线全身照射后,胸腺细胞内细胞凋亡相关基因p53、Bcl-2和Bax蛋白的表达进行了比较研究。结果表明,75mGyX射线全身照射后24h,p53基因蛋白产物的表达显著降低(P<0.005),而2GyX射线全身照射后24h,p53基因蛋白表达则显著增高(P<0.005)。75mGyX射线全身照射后24h,Bcl-2/Bax比值明显增加(P<0.05)。而2GyX射线全身照射组,Bcl-2/Bax比值无明显变化。提示:p53和Bcl-2/Bax在辐射诱导的胸腺细胞凋亡中起着重要作用。
The expressions of p53, Bcl-2 and Bax proteins in thymocytes were compared using flow cytometry after 75 mGy and 2Gy X-ray irradiation in Kunming mice. The results showed that the expression of p53 gene protein was significantly decreased (P <0.005) 24h after 75mGy X - ray irradiation, while p53 gene protein expression was significantly increased 24 h after 2GYX irradiation (P <0.005). The ratio of Bcl-2 / Bax was significantly increased at 24 h after 75 mGy X-ray irradiation (P <0.05). However, there was no significant change in Bcl-2 / Bax ratio in 2Gy X-ray whole body irradiation group. Tip: p53 and Bcl-2 / Bax play an important role in radiation-induced thymocyte apoptosis.
