本研究以宁夏枸杞为材料, 根据GenBank上已经登录的该基因的保守区设计简并引物, 通过RTPCR扩增获得基因保守片段, 利用RACE技术获得该基因3'端序列.最终, 经拼接后得到一个长约1 802 bp的ms2基因片段 (登录号为JQ341412.1), 终止密码子为TAG, 3'端包含非编码区和Poly A尾巴.相应编码基因片段527个氨基酸的肽段.生物信息学分析表明, ms2基因序列中含有NAD (P) H区域, 该区域含有显著的TGXXGXXG结构单元, 以及微体结合信号区域.从第44个氨基酸到第346个氨基酸为NAD结合区, 从第459个氨基酸到第517个氨基酸为雄性不育C-末端区.同源性检索表明, ms2基因片段氨基酸序列与亚麻、葡萄、大豆、拟南芥和青菜的同源性分别为71%、71%、72%、65%和66%.系统进化树分析表明, 宁夏枸杞ms2基因与葡萄的亲缘关系最近, 与毛果杨、亚麻以及十字花科的植物聚为一类, 而与江南卷柏的亲缘关系最远.RT-PCR分析表明, ms2基因在可育花蕾的2、3时期表达非常强, 而在不育花蕾的2、3时期表达较弱.“,”Specific primers were de signed according to the conserved sequences of ms2 genes in Gen Bank and fragment of ms2 genes specific to male sterility were obtained by amplifying genomic DNA of the male fertile line of Lycium barbarum L. the primers, and 3'c DNA was cloned using RACE technology. The fragment of the gene was 1 808 bp in length by joint, which was submitted to Gen Bank under accession number JQ341412. 1, contained the ending code-TAG, the fragment of 3'-terminal and Poly A tail. Its corresponding encoded 527 amino acids. In bio-information analysis, the corresponding peptide segment contained NAD (P) H-binding protein. It typically had a TGXXGXXG cofactor binding motif and microbody targeting signal. There was NAD (P) H-binding from 44 amino acid to 346 and N-terminal of male sterility from 459 amino acid to 517. Through Blastp program, the ms2 protein from Lycium barbarum L. were found sharing 71%, 71%, 72%, 65%, 66% identity with the known ms2 proteins from Linum usitatissimum, Vitis vinifera, Glycine max, Arabidopsis thaliana and Brassica chinensis, respectively. In phylogenetic analysis, the ms2 gene had closely relativeship with grape, which was clustersed in a group with Populus trichocarpa, Linum usitatissimum and Cruciferae, and had distantly relativeship with Moellendorf's Spikemoss Herb. RT-PCR revealed that the ms2 gene expressed strongly in flower buds of stage Ⅱ and stage Ⅲ of the male fertile lines, while expressed weakly in flower buds of stage Ⅱ and stage Ⅲ from the male sterility lines.
