目的:建立超高效液相色谱-四极杆飞行时间质谱(UPLC-Q-TOF-MS)对罗布麻叶中黄酮类成分分析的方法。方法:采用Alltima C18色谱柱(250 mm×4.6 mm,5μm),以乙腈-1%醋酸为流动相,梯度洗脱,流速为0.8mL/min,检测波长为254 nm,柱温为30℃。在电喷雾负离子模式下监测采集数据。结果:初步鉴定出11个黄酮类化合物,并同时测定其中6个黄酮的含量。6个黄酮的浓度与峰面积的线性关系良好(r>0.9996),加样回收率为94.3%~103.3%。结论:该方法准确可靠,重现性好,可作为罗布麻叶内在质量评价的依据。
Objective: To establish a method for the determination of flavonoids in Apocynum venetum L. by UPLC-Q-TOF-MS. Methods: Alltima C18 column (250 mm × 4.6 mm, 5 μm) was used with mobile phase of acetonitrile-1% acetic acid as the mobile phase. The flow rate was 0.8 mL / min. The detection wavelength was 254 nm and the column temperature was 30 ℃. Acquisition data monitored in electrospray negative ion mode. Results: Eleven flavonoids were initially identified and six flavonoids were simultaneously assayed. The linear relationship between the concentration of 6 flavonoids and the peak area was good (r> 0.9996), and the recoveries were 94.3% -103.3%. Conclusion: The method is accurate and reliable, reproducible, and can be used as the basis for evaluating the intrinsic quality of Apocynum venetum leaves.
