目的:研究活血利湿方对非酒精性脂肪性肝病(Non-alcoholic fatty liver disease,NAFLD)大鼠肝功能及肝脏病理的影响。方法:利用高脂饲料建立SD大鼠非酒精性脂肪肝模型,期间预防组大鼠予活血利湿方灌胃,8周造模成功后,取血及肝脏,观察活血利湿方的预防效果。造模成功大鼠分为三组,分别予活血利湿方水煎剂、荷丹片溶液、生理盐水4周后,取血及肝脏。两批大鼠均检测血清中谷丙转氨酶(ALT)、天门冬氨酸氨基转移酶(AST)、乳酸脱氢酶(LDH)水平,同时观察大鼠肝组织病理学形态的变化,评估NAFLD活动度积分(NAFLD activity score,NAS)及炎症分级。结果:与空白组相比,模型组大鼠ALT水平不变(P>0.05),AST、LDH升高(P<0.05),预防组AST、LDH较模型组降低。与模型组相比,活血利湿方组NAS总积分及炎症分级均下降(P<0.05),有统计学差异。结论:活血利湿方可改善非酒精性脂肪肝大鼠肝功能、肝脏病理形态,减轻炎症程度。
Objective: To investigate the effects of Huoxue Lishi recipe on liver function and liver pathology in rats with non-alcoholic fatty liver disease (NAFLD). Methods: The non-alcoholic fatty liver model of SD rats was established by using high-fat diet. During the period of prophylaxis, the rats in the prophylaxis group were given gavage of dampness and gavage. After 8 weeks of successful model establishment, blood and liver were taken for observation of the preventive effect of Huoxue-dampness prescription. The successful rats were divided into three groups, respectively, to take blood and dampness decoction, Haldine tablets solution, saline for 4 weeks, blood and liver. Serum levels of alanine aminotransferase (ALT), aspartate aminotransferase (AST) and lactate dehydrogenase (LDH) in both batches of rats were measured. The pathological changes in rat liver tissue were also observed to assess the activity of NAFLD Score (NAFLD activity score, NAS) and inflammation grade. Results: Compared with the blank group, the levels of ALT and AST in model group were unchanged (P> 0.05), AST and LDH were increased (P <0.05), and the levels of AST and LDH in prevention group were lower than those in model group. Compared with the model group, the total score of NAS and the grade of inflammation in Huoxue Lishi decoction decreased (P <0.05), with statistical difference. Conclusion: Huoxue and dampness prescription can improve liver function, liver pathology and alleviate inflammation in non-alcoholic fatty liver disease rats.
