颐脑解郁方对脑出血后抑郁大鼠脑内5-羟色胺和5-羟色胺1A受体表达的影响

目的:探讨脑出血后抑郁动物模型脑内单胺神经递质5-羟色胺(5-HT)、5-羟色胺1A受体(5-HT1AR)的变化,研究中药颐脑解郁方对脑出血后抑郁的干预机理。方法::用免疫组织化学方法观察脑出血后抑郁模型大鼠脑内5-HT,5-HT1AR的表达变化及以西药百忧解为对照,中药颐脑解郁方的干预作用。结果:脑出血后抑郁模型大鼠脑海马CA3、扣带皮质、隔核内5-HT阳性细胞表达减少,颐脑解郁方和百忧解均增加海马CA3、扣带皮质内5-HT阳性细胞内蛋白表达,减少5-HT阳性细胞在下丘脑表达;模型组大鼠海马CA3、隔核、下丘脑5-HT1AR阳性细胞表达较正常增加,百忧解和颐脑解郁方均可增加海马CA3区、扣带皮质5-HT1AR阳性细胞表达,但百忧解减少隔核、下丘脑内5-HT1AR阳性细胞表达,颐脑解郁方减少下丘脑内5-HT1AR阳性细胞表达。结论:中药颐脑解郁方可以通过增强5-HT阳性细胞的蛋白表达,调节不同部位5-HT1AR细胞功能起到改善抑郁的作用。 Objective: To investigate the changes of monoamine neurotransmitters 5-hydroxytryptamine (5-HT) and 5-hydroxytryptamine 1A receptor (5-HT1AR) in the brain following depressive cerebral hemorrhage and to investigate the effect of Chinese herbal compound Xiaoyu Jieyu Fang on depression after cerebral hemorrhage Intervention mechanism. Methods: Immunohistochemistry was used to observe the expression of 5-HT, 5-HT1AR in the brains of rats with depression after intracerebral hemorrhage. The interventional effect of the Chinese herbal compound Xiaoyu Decoction was compared with the western drug Prozac. Results: The expression of 5-HT-positive cells in hippocampal CA3, cingulate cortex, and septal nucleus decreased after cerebral hemorrhagic depression in rats with cerebral hemorrhage. Both Qiao Jieyu Decoction and Prozac solution increased 5-HT-positive cells in hippocampal CA3 and cingulate cortex. Protein expression decreased expression of 5-HT positive cells in the hypothalamus; expression of 5-HT1AR positive cells in the hippocampus CA3, septal nucleus and hypothalamus was increased in the model group compared with the normal group. Both Pajahide and Camphor Yujie can increase the hippocampal CA3 area and deduction. 5-HT1AR positive cells were expressed in the cortex, but Prozac reduced the expression of 5-HT1AR positive cells in the nucleus pulposus and hypothalamus. The Qiao Jieyu Decoction decreased the expression of 5-HT1AR positive cells in the hypothalamus. Conclusion: The Chinese herbal group Xiaoyu Jieyu Fang can improve the function of 5-HT1AR cells by enhancing the expression of 5-HT-positive cells and regulating the function of 5-HT1AR cells in different parts.
