实验选择了不同浓度的rhG-CSF(0.015~0.50μg/ml)体外作用于HL-60细胞5天,检测其细胞数、~3H-TdR掺入量和细胞内cAMP水平。结果表明,细胞数和~3H-TdR掺入量都随rhG-CSF浓度增加而明显下降。在浓度达0.125μg/ml时,这种抑制作用基本达坪值;而细胞内cAMP水平随rhG-CSF浓度增加明显增高,呈剂量依赖性。推测rhG-CSF是通过诱导细胞分化来实现对HL-60细胞增殖的抑制作用。
Different concentrations of rhG-CSF (0.015-0.50 μg/ml) were used to treat HL-60 cells in vitro for 5 days. The number of cells, ~3H-TdR incorporation, and intracellular cAMP levels were measured. The results showed that both the number of cells and the amount of ~3H-TdR incorporated decreased with the increase of rhG-CSF concentration. At a concentration of 0.125 μg/ml, this inhibition reached a plateau, and intracellular cAMP levels increased significantly with increasing concentrations of rhG-CSF in a dose-dependent manner. It is speculated that rhG-CSF inhibits the proliferation of HL-60 cells by inducing cell differentiation.
