目的研究程序性死亡分子配体在HepG2和2.2.15细胞表面的表达及其与HBV感染的关系。方法2007年3月至7月于北京大学第一医院分别培养HepG2细胞和2.2.15细胞,用荧光素标记的抗程序性细胞死亡分子配体-1、2(PD-L1、PD-L2)特异性抗体标记,行流式细胞仪检测;同时提取细胞核糖核酸(RNA),用逆转录聚合酶链反应(RT-PCR)检测PD-L1和PD-L2mRNA的表达。结果PD-L1在2.2.15细胞的表达显著上调;无论是膜蛋白质或是mRNA形式,PD-L2在HepG2细胞和2.2.15细胞均不表达。结论HBV感染导致肝细胞表面程序性死亡分子的配体表达上调,提示与病毒免疫逃避有关。
Objective To study the expression of apoptosis ligand on HepG2 and 2.2.15 cells and its relationship with HBV infection. Methods HepG2 cells and 2.2.15 cells were cultured in Peking University First Hospital from March 2007 to July 2007 respectively. Fluorescein-labeled anti-apoptotic ligand-1,2 (PD-L1, PD-L2) Specific antibody was detected by flow cytometry. Meanwhile, RNA was extracted and the expression of PD-L1 and PD-L2 mRNA was detected by reverse transcription-polymerase chain reaction (RT-PCR). Results The expression of PD-L1 in 2.2.15 cells was significantly up-regulated. PD-L2 was not expressed in both HepG2 and 2.2.15 cells, both in membrane protein and mRNA. Conclusion HBV infection leads to the up-regulation of ligand expression on programmed cell death in hepatocytes, suggesting that it is related to virus immune evasion.
