以青花菜(Brassica oleracea L.var.italica Plenck)‘B908’和‘B1092’为材料,通过荧光染色技术对经25、4、32.5和(4→32.5)℃等不同温度预处理的小孢子的分裂方式进行观察,并对出胚率进行统计。结果表明,不同温度预处理的小孢子胚胎发生途径基本相同,均以对称分裂产生胚胎的孢子体发育途径为主(B途径),经历球形胚、心形胚、鱼雷胚,最终形成子叶形胚状体。但是不同温度预处理的小孢子启动胚胎发生的频率不同,(4→32.5)℃预处理后的成胚率最高。
The microspores pretreated with different temperatures of 25, 4, 32.5 and (4 → 32.5) ℃ were obtained by fluorescence staining using Brassica oleracea L.var.italica Plenck, ’B908’ and ’B1092’ Split way to observe, and out of rate statistics. The results showed that the pathways of microspore embryo pretreatment at different temperatures were basically the same, both of which were dominated by symmetrical cleavage of embryos (B pathway). The embryos underwent globular embryo, cardioid embryo, torpedo embryo and finally cotyledon embryo Body. However, the frequency of embryogenesis of microspores with different temperature pretreatment was different, and the embryogenesis rate was highest after (4 → 32.5) ℃ pretreatment.
