目的:研究四逆散对海马神经元发生的影响。方法:采用皮质酮(CORT)抑制海马神经元发生的体外模型,在原代海马神经元培养体系中加入CORT后,运用MTT法测定神经元存活率,以5-溴脱氧尿嘧啶核苷(Brdu)标记增殖细胞,以神经元特异性烯醇化酶(NSE)标记成熟神经元生长。结果:四逆散含药血清能够提高CORT作用下海马神经元的存活率(P<0.05),增加了培养海马神经元Brdu数目(P<0.01)和NSE阳性细胞数量(P<0.05),并能促进突起生长。结论:四逆散含药血清可以有效地支持高皮质激素环境中神经元存活、增殖、生长,具有一定的促进海马神经元发生的作用。四逆散的抗抑郁作用与此机制有关。
Objective: To study the effect of Sini Decoction on the neurons in hippocampus. Methods: CORT was used to inhibit the hippocampal neurons in vitro.
