本实验是把显示葡萄糖-6-磷酸酶(G-6-Pase)的电镜细胞化学技术应用于研究急性砷中毒大鼠心肌G-6-Pase活性的变化,目的在于从亚显微水平上进一步探讨砷对心肌结构和功能的影响。选用健康wistar大鼠20只,随机分成实验组和对照组,每组10只。实验组大鼠每天自由饮用含As_2O_350PPm的蒸馏水,对照组饮用蒸馏水。35天后急性处死取其心尖部组织,置于4%多聚甲醛+1%戊二醛内前固定,入孵育液,37℃下孵育60min,1%锇酸后固定,常规脱水包埋,超薄切片,不经电子染色进行电镜观察。
In this study, we used the electron microscopy cytochemistry technique to display G-6-Pase in the study of myocardial G-6-Pase activity in acute arsenic poisoning rats.
