目的:探讨内毒素诱导肝损害家兔血浆和肝组织中肿瘤坏死因子α(TNFα)、白细胞介素8(IL8)水平变化及热毒清的保肝作用。方法:采用ELISA法检测内毒素性全身性Shwartzman反应家兔(作为播散性血管内凝血肝损害模型)血浆和肝组织匀浆中TNFα、IL8水平,同时观察谷丙转氨酶(ALT)、酸性磷酸酶(ACP)活性变化及热毒清对它们的影响。结果:给家兔注射内毒素后血浆及肝组织内TNFα、IL8水平显著升高,二者呈良好的线性正相关,血浆IL8水平与酶活性变化及肝损害程度密切相关;热毒清能明显抑制TNFα、IL8的产生,并能减轻肝损害程度。结论:热毒清对TNFα、IL8活性的抑制作用是热毒清时保护肝细胞的重要机制。
Objective: To investigate the changes of tumor necrosis factor α (TNFα) and interleukin 8 (IL8) levels in liver and plasma of hepatic injury induced by endotoxin and the hepatoprotective effect of JTD. Methods: The levels of TNFα and IL8 in plasma and liver homogenate of endotoxin-induced Shwartzman reaction rabbits (as a model of disseminated intrahepatic coagulation damage) were detected by ELISA. Meanwhile, the effects of alanine aminotransferase (ALT), acid phosphatase Changes in enzyme (ACP) activity and the effect of heat detoxification on them. Results: The levels of TNFα and IL8 in plasma and liver tissue of rabbits after injection of endotoxin were significantly increased, which showed a good linear correlation. The level of plasma IL8 was closely related to the changes of enzyme activity and the degree of liver damage. Inhibit TNFα, IL8 production, and reduce the extent of liver damage. CONCLUSION: The inhibitory effect of TLP on TNFα and IL8 activity is an important mechanism of protecting hepatocytes when DHT is treated.
