阴茎勃起功能障碍(erectile dysfunction,ED)是影响男性健康的一种常见疾病,其患病率大约是20%~45%。近年来,ED的治疗取得了很大的进展,尤其是高选择性磷酸二酯酶5型(phosphodiesterase V,PDE_5)抑制剂西地那非的问世,使ED的治疗发生了根本性的改变。然而选择性PDE_5抑制剂需长期服药,治疗费用昂贵,加之头痛、面部潮红、消化不良、鼻炎、视觉异常等副作用,从而限制了临床上的应用。近年来,基因治疗ED的研究取得了一些可喜的成果,展示了美好的前景。ED的基因治疗有很多优势:(1)阴茎是体表器官,能准确定位;(2)治疗时在阴茎根部扎止血带,能局限目的基因,减少不良反应;(3)阴茎海绵体细胞间存在缝隙连接,少量细胞转染目的基因即可有效;(4)阴茎平滑肌细胞更新较慢,可使目
Erectile dysfunction (erectile dysfunction, ED) is a common disease affecting men’s health, the prevalence is about 20% to 45%. In recent years, tremendous progress has been made in the treatment of ED. In particular, the advent of sildenafil, a highly selective phosphodiesterase V (PDE_5) inhibitor, has fundamentally changed the treatment of ED. However, selective PDE_5 inhibitors require long-term medication, expensive treatment, coupled with headaches, facial flushing, indigestion, rhinitis, visual acuity and other side effects, thus limiting the clinical application. In recent years, gene therapy ED research has made some gratifying achievements, showing a bright future. ED gene therapy has many advantages: (1) the penis is the body surface organ, can accurately locate; (2) the treatment of penile tourniquet at the root of the penis, can limit the purpose of gene, reduce adverse reactions; (3) The existence of gap junctions, a small amount of cells transfected with the purpose of the gene can be effective; (4) penile smooth muscle cell renewal is slower,
