目的探讨早期糖尿病膀胱舒张功能及肾上腺素能β3受体的改变的分子生物学意义。方法将新生3d的Wistar雌性大鼠随机分为2组,各30只,实验组以1%链尿佐菌素诱导为糖尿病大鼠,比较2组大鼠离体全膀胱舒张压力变化、β3受体及cAMP含量改变。结果2、4、12周时,糖尿病组膀胱舒张压力分别为(11.8±2.3)、(11.9±2.2)、(11.5±3.0)cmH2O,对照组为(32.8±7.6)、(33.9±5.2)、(34.7±2.7)cmH2O;WestonBlot显示2、4、12周时,糖尿病组膀胱逼尿肌β3受体灰度值分别为(104±7)、(105±11)、(126±14)μm,对照组为(151±11)、(151±10)、(152±13)μm。放免检测显示2、4、12周时,糖尿病组膀胱逼尿肌细胞中cAMP含量分别为(0.195±0.026)、(0.198±0.030)、(0.109±0.031)pmol/L;对照组为(0.349±0.051)、(0.350±0.052)、(0.353±0.031)pmol/L。2组比较差异均有统计学意义(P<0.05)。结论糖尿病早期膀胱逼尿肌舒张功能受损,支配逼尿肌舒张功能的肾上腺素能β3受体及受体后生化途径发生异常。
Objective To investigate the molecular biological significance of bladder diastolic function and adrenergic β3 receptor in early stage of diabetes mellitus. Methods Newborn 3d female Wistar rats were randomly divided into two groups, 30 rats in each group. The rats in experimental group were induced with 1% streptozotocin to induce diabetic rats. Comparing the changes of isolated bladder diastolic pressure in vitro, Body and cAMP content changes. Results The diastolic pressure of bladder in diabetic group was (11.8 ± 2.3), (11.9 ± 2.2) and (11.5 ± 3.0) cmH2O at 2, 4 and 12 weeks respectively, and was 32.8 ± 7.6 and 33.9 ± 5.2 in control group (34.7 ± 2.7) cmH2O, respectively. WestonBlot showed that the gray value of β3 receptor in the detrusor of diabetic rats were (104 ± 7), (105 ± 11) and (126 ± 14) The control group was (151 ± 11), (151 ± 10), (152 ± 13) μm. Radioimmunoassay showed that the cAMP contents in detrusor cells of diabetic bladder were (0.195 ± 0.026), (0.198 ± 0.030) and (0.109 ± 0.031) pmol / L at 2, 4 and 12 weeks respectively 0.051), (0.350 ± 0.052), (0.353 ± 0.031) pmol / L. The differences between the two groups were statistically significant (P <0.05). Conclusions In early stage of diabetes, the diastolic function of detrusor muscle is impaired. The adrenergic β 3 receptor which controls detrusor diastolic function and the post-receptor biochemical pathway are abnormal.
