目的:观察尼氟灭酸(NFA)对增殖的大鼠气道平滑肌细胞(3-5代ASMC)原癌基因c-fos表达的影响。方法:分离大鼠气道平滑肌细胞,放入含15%胎牛血清的培养基中培养,第35代细胞用于实验。向细胞中加入不同浓度NFA(0、10、50、100μmol/L),分别作用1、2、3h后,收集细胞,提取总RNA,通过逆转录-聚合酶链反应(RT-PCR)的方法检测原癌基因c-fos的表达。结果:高、中、低浓度的NFA组气道平滑肌细胞中c-fos基因的表达水平在3个时间点均明显低于对照组,且随着NFA浓度的增高,表达量逐渐减少。结论:NFA对增殖的大鼠气道平滑肌细胞c-fos基因的表达有明显的抑制作用。
Objective: To observe the effect of niflumic acid (NFA) on the expression of proto-oncogene c-fos in cultured rat airway smooth muscle cells (3-5 passages ASMC). Methods: Rat airway smooth muscle cells were isolated and cultured in medium containing 15% fetal bovine serum. The 35th generation cells were used in the experiment. The cells were treated with different concentrations of NFA (0, 10, 50 and 100 μmol / L) for 1, 2 and 3 h, respectively. Cells were harvested and the total RNA was extracted by reverse transcription polymerase chain reaction Detection of proto-oncogene c-fos expression. Results: The expression of c-fos gene in NFA group was significantly lower than that in control group at three time points, and the expression decreased gradually with the increase of NFA concentration. Conclusion: NFA can significantly inhibit the expression of c-fos gene in airway smooth muscle cells of rats.
