目的 观察缺氧条件下培养的肺微血管内皮细胞 (PMEC)培养液对大鼠主动脉平滑肌细胞(ASMC)的作用。方法 应用倒置显微镜和电镜观察细胞形态 ,同位素放射测定3H 胸腺嘧啶核苷 ( 3H TdR)摄取量 ,流式细胞仪测定细胞的周期和蛋白含量。结果 单纯缺氧可使ASMCG0 G1 期数目增多。3H TdR摄取量降低 ,蛋白质含量减少 ;缺氧的PMEC培养液能促进大鼠ASMC从G0 G1 期进入S期 ,3H TdR含量升高 ,使蛋白质含量进一步降低。结论 缺氧可能刺激微血管的内皮细胞分泌某些活性物质 ,进而促进动脉平滑肌细胞增生。
Objective To observe the effects of cultured pulmonary microvascular endothelial cells (PMECs) cultured in hypoxia on rat aortic smooth muscle cells (ASMCs). Methods The morphological changes of the cells were observed by inverted microscope and electron microscope. 3H - thymidine uptake by 3H - thymidine (3H) was measured by radioisotope assay. Cell cycle and protein content were determined by flow cytometry. Results Hypoxia alone increased the number of G1 phase of ASMCG0. 3H TdR uptake decreased and protein content decreased. Hypoxic PMEC culture medium promoted the progression of ASMC from G0 G1 phase to S phase, and the 3H TdR content increased and the protein content decreased further. Conclusions Hypoxia may stimulate the endothelial cells of microvessels to secrete some active substances, and then promote the proliferation of arterial smooth muscle cells.
