目的:探讨天合保肾合剂对糖尿病肾病(DN)大鼠血管内皮功能的影响。方法:大鼠经背部切除单侧肾,2周后以链尿佐菌素(STZ)尾静脉注射诱导DN模型,观察天合保肾合剂高、中、低剂量组对DN大鼠肾组织血管内皮生长因子(VEGF)、内皮素(ET)、血小板活性指标血浆血栓素B2(TXB2)与6-酮-前列腺F1α(6-K-PGF1α)含量影响。结果:天合保肾合剂各剂量组均能够降低DN大鼠肾组织VEGF、ET、血浆TXB2水平,升高血浆6-K-PGF1α水平,与模型对照组比较有显著性差异(P<0.01),并呈剂量依赖性。结论:天合保肾合剂能够降低肾组织VEGF、ET水平及TXB2/6-K-PGF1α比值,改善DN大鼠的血管内皮功能障碍。
Objective: To investigate the effect of Tianhe Baoshen mixture on the vascular endothelial function in diabetic nephropathy (DN) rats. METHODS: Rats were treated with unilateral nephrectomy. Two weeks later, DN model was induced by tail vein injection of streptozotocin (STZ). The blood vessels of kidneys in DN rats were observed with high, medium and low doses of Tianhe Baoshen mixture. The levels of serum thromboxane B2 (TXB2) and 6-keto-prostate F1α (6-K-PGF1α) were affected by endothelial growth factor (VEGF), endothelin (ET) and platelet activity. Results: Each dose of Tianhe Baoshen mixture could reduce the level of VEGF, ET and plasma TXB2 in kidney tissue of DN rats, and increase the level of plasma 6-K-PGF1α, which was significantly different from the model control group (P<0.01). , and in a dose-dependent manner. Conclusion: Tianhe Baoshen Mixture can reduce the level of VEGF, ET and the ratio of TXB2/6-K-PGF1α in renal tissue and improve the vascular endothelial dysfunction in DN rats.
