目的:观察辣椒素对人大细胞肺癌细胞NCI-H460侵袭能力及E钙黏蛋白(E-cadherin)蛋白,Slug蛋白表达的影响,探讨其抗非小细胞肺癌的可能机制。方法:体外培养NCI-H460细胞,以不同终浓度辣椒素进行处理,未加辣椒素为空白组。采用MTT比色分析法,观察辣椒素对NCI-H460细胞增殖抑制作用,计算半数抑制浓度(IC50);采用微丝荧光染色和Transwell小室细胞侵袭实验观察NCI-H460细胞侵袭能力,以及Western blot和RT-PCR分析检测E-cadherin和Slug蛋白及mRNA表达的变化。结果:MTT比色分析法结果显示,辣椒素对NCI-H460细胞生长具有明显抑制作用,与空白组比较,差异具有统计学意义(P<0.05);微丝荧光染色分析表明,辣椒素可明显抑制NCI-H460细胞丝状伪足形成;Transwell体外侵袭实验结果显示,辣椒素可显著抑制侵袭穿膜细胞数,与空白组比较,差异具有统计学意义(P<0.05)。Western blot和RT-PCR分析结果显示,辣椒素显著升高E-cadherin表达水平,并显著降低Slug表达水平,与空白组比较,差异具有统计学意义(P<0.05)。结论:辣椒素能明显抑制NCI-H460细胞活率,降低侵袭能力与其降低Slug表达,增加E-cadherin表达有关,可能是其抗非小细胞肺癌的机制之一。
Objective: To investigate the effect of capsaicin on the invasiveness of human large cell lung cancer cell line NCI-H460 and the expression of E-cadherin and Slug proteins and to explore the possible mechanism of anti-non-small cell lung cancer. Methods: NCI-H460 cells were cultured in vitro and treated with different concentrations of capsaicin. No capsaicin was added as a blank group. MTT assay was used to observe the inhibitory effect of capsaicin on the proliferation of NCI-H460 cells, and the half inhibitory concentration (IC50) was calculated. The invasion ability of NCI-H460 cells was observed by means of microfilament fluorescence staining and transwell cell invasion assay. RT-PCR analysis of E-cadherin and Slug protein and mRNA expression changes. Results: MTT colorimetric assay showed that capsaicin significantly inhibited the growth of NCI-H460 cells. Compared with the blank group, the difference was statistically significant (P <0.05). The results of microfilament fluorescence staining showed that capsaicin can be significantly Inhibit the formation of filopodia in NCI-H460 cells. The results of transwell invasion assay showed that capsaicin can significantly inhibit the number of invasive transmembrane cells, compared with the blank group, the difference was statistically significant (P <0.05). Western blot and RT-PCR analysis showed that capsaicin significantly increased E-cadherin expression and significantly decreased Slug expression compared with the blank group, the difference was statistically significant (P <0.05). Conclusion: Capsaicin can significantly inhibit the survival rate of NCI-H460 cells and reduce its invasion ability, which may be related to the decrease of Slug expression and E-cadherin expression, which may be one of the mechanisms of its anti-non-small cell lung cancer.
