目的:研究黔药缬草挥发油的化学成分及其对小鼠抗炎镇痛作用。方法:采用超临界CO2技术提取缬草挥发油,采用气相色谱-质谱(GC-MS)联用仪分析缬草挥发油的化学成分。采用二甲苯致小鼠耳肿胀试验、冰醋酸致小鼠扭体试验对缬草挥发油进行抗炎镇痛活性评价。结果:从缬草中分离鉴定了52个化合物,占挥发油总含量的92.558%。与模型组比较,缬草挥发油高剂量组能显著抑制二甲苯致小鼠耳廓肿胀(P<0.05),显著减少冰醋酸致小鼠扭体次数(P<0.01)。结论:GC-MS法可以作为缬草药材质量标准的分析方法。缬草挥发油具有较好的抗炎镇痛作用,可为缬草的开发应用研究提供参考。
Objective: To study the chemical constituents of the volatile oil of Valerian in Guizhou and its anti-inflammatory and analgesic effects on mice. Methods: Volatile oil from Valeriana officinalis was extracted by supercritical CO2. The chemical constituents of Valerian volatile oil were analyzed by gas chromatography-mass spectrometry (GC-MS). The anti-inflammatory and analgesic activities of valerian volatile oil were evaluated by xylene-induced mouse ear swelling test and acetic acid writhing test. Results: Fifty-two compounds were isolated and identified from Valerian, accounting for 92.558% of the total volatile oil. Compared with the model group, the valerian oil high-dose group can significantly inhibit xylene-induced ear swelling in mice (P <0.05), significantly reduce the number of writhing caused by glacial acetic acid (P <0.01). Conclusion: GC-MS method can be used as an analytical method for the quality standard of valerian herbs. Valerian volatile oil has good anti-inflammatory analgesic effect, which can provide reference for the development and application of Valerian.
