目的研究菘蓝有效成分在体内外抗甲型流感病毒的作用。方法体外用MTT法检测菘蓝有效成分对传代狗肾细胞(MDCK)的毒性浓度,观察菘蓝有效成分抑制甲型流感病毒的致细胞病变作用(CPE),MTT法检测菘蓝有效成分对流感病毒的抑制率;建立在体流感病毒感染小鼠肺炎模型,通过观察小鼠肺病变程度指标,测定肺组织内流感病毒血凝滴度判定菘蓝有效成分对小鼠肺炎的治疗作用。结果菘蓝有效成分在体外主要抑制甲型流感病毒在细胞内的复制,其对病毒的半数抑制率浓度为64μg.mL-1,治疗指数达28.88;口服菘蓝有效成分能明显减轻小鼠肺组织病变程度,对肺组织内病毒的增殖有明显抑制作用。结论菘蓝有效成分具有较好的抗甲型流感病毒的作用,且呈剂量效应关系。
Objective To study the effect of the active ingredients of radix islaniridae against influenza A virus in vitro and in vivo. Methods MTT assay was used to detect the cytotoxicity of effective components of Isatis indigo to MDCK. The inhibitory effect of active ingredients of Isatis indigotica on the cytopathic effect (CPE) of influenza A virus was observed by MTT assay. Virus inhibition rate; established in the mouse model of influenza-infected mice pneumonia, by observing the degree of lung lesions in mice, determination of pulmonary blood clotting titer of influenza virus to determine the effective components of the treatment of mouse pneumonia in mice. Results The main constituents of the extract were mainly inhibit the replication of influenza A virus in vitro. The half inhibitory rate against virus was 64 μg.mL-1 and the therapeutic index was 28.88. The degree of tissue lesions, lung tissue proliferation of the virus was significantly inhibited. Conclusion The effective components of Isatis have better anti-influenza A virus effect, and the dose-response relationship.
