目的:探讨甲氧滴滴涕(MXC)对绒毛外滋养细胞(EVCT)白细胞表面抗原G(HLA-G)表达的影响。方法:用胰蛋白酶消化加流式细胞仪分离得到人绒毛外滋养细胞,进行原代培养及鉴定。将所获细胞分为MXC(1μmol/L、3μmol/L、5μmol/L、7μmol/L)处理组及空白对照组,培养12h、24h、48h。用Western blotting检测各组各时间点HLA-G的表达,同时用RT-PCR法检测HLA-GmRNA表达。结果:MXC处理组滋养细胞HLA-G表达呈时间剂量依赖性下降,HLA-G mRNA变化与其一致。结论:MXC可从蛋白和mRNA水平降低绒毛外滋养细胞HLA-G表达,从而降低其免疫耐受功能。
Objective: To investigate the effect of Methoxy-DDT (MXC) on the expression of leukocyte surface antigen G (HLA-G) in extravillous trophoblast cells (EVCT). Methods: Human chorionic trophoblast cells were isolated by trypsin digestion and flow cytometry, and were primary cultured and identified. The cells were divided into MXC (1μmol / L, 3μmol / L, 5μmol / L, 7μmol / L) treatment group and blank control group, cultured for 12h, 24h, 48h. The expression of HLA-G was detected by Western blotting at each time point, and the expression of HLA-G mRNA was detected by RT-PCR. Results: The expression of HLA-G in trophoblast cells in MXC treatment group decreased in a dose-and time-dependent manner, and the changes of HLA-G mRNA were consistent with those in MXC treated group. Conclusion: MXC can reduce the expression of HLA-G in extravillous chondrocytes from the level of protein and mRNA, thereby reducing its immune tolerance function.
