目的 探讨小鼠髓系DC(CD8α-)中4-1BB的功能性表达。方法 采用免疫荧光标记和流式细胞仪检测DC上4-1BB分子的表达,用酶联免疫方法检测DC分泌的IL-2和IL-12。结果 小鼠髓系DC前体细胞中已有4-1BB分子表达,且随着DC的分化发育和成熟,其表达上调。激发型抗4-1BB分子的抗体2A刺激的DC分泌IL-2和IL-12能力增强,而2A抗体联合TNF-α刺激的DC分泌IL-2和IL-12的量明显高于两者单独刺激组(P<0.05)。结论 小鼠髓系DC表达的4-1BB具有功能性。
Objective To investigate the functional expression of 4-1BB in murine myeloid DC (CD8α-). Methods The expression of 4-1BB in DCs was detected by immunofluorescence staining and flow cytometry. The levels of IL-2 and IL-12 secreted by DCs were detected by enzyme-linked immunosorbent assay. Results 4-1BB molecules were expressed in mouse myeloid DC precursor cells, and their expression was up-regulated as DCs differentiated. The level of IL-2 and IL-12 secreted by the stimulated anti-4-1BB antibody-stimulated DCs was increased, while the levels of IL-2 and IL-12 secreted by the 2A antibody combined with TNF-α-stimulated DCs were significantly higher than those of the two anti- Stimulation group (P <0.05). Conclusion 4-1BB expressed in mouse myeloid DC is functional.
