以灵芝菌丝体为实验材料,采用单因素分组实验法对胞内多糖提取工艺(水提法、碱提法和超声提取法)进行了初步探讨。确定超声波提取法是菌丝体胞内多糖提取的最高效方法,其最佳提取条件为:料液比为1:30,提取温度70℃,超声功率为400W,提取时间20min,多糖提取率为3.36%。超声波提取法与水提法、碱提法相比,提取时间缩短5/6,提取温度降低20℃,提取效率分别高出44.8%和11.3%。
Using Ganoderma lucidum mycelium as experimental material, the extraction process of intracellular polysaccharides (water extraction, alkali extraction and ultrasonic extraction) was studied by single factor grouping method. Ultrasonic extraction was the most efficient method for the extraction of intracellular polysaccharides from mycelia. The optimal extraction conditions were as follows: the ratio of solid to liquid was 1:30, the extraction temperature was 70 ℃, the ultrasonic power was 400W, the extraction time was 20min, the extraction rate of polysaccharide was 3.36%. Compared with water extraction method and alkali extraction method, the extraction time was shortened by 5/6 and the extraction temperature was decreased by 20 ℃. The extraction efficiency was 44.8% and 11.3% higher than that by ultrasonic extraction.
